Improved generalized transducing bacteriophage for Caulobacter crescentus.

چکیده
Caulobacter phage phi Cr30T, a temperature-sensitive derivative of the lytic, generalized transducing phage phi Cr30, was isolated as a double temperature-sensitive recombinant in a cross between phi Cr30ts1 and phi Cr30ts2, phi Cr30T mediated generalized transduction of Caulobacter crescentus at frequencies comparable to those of phi Cr30 and eliminated the requirement for irradiation of transducing lysates to prevent killing of transductants on the plate.

Generalized Transduction in CAULOBACTER CRESCENTUS.
Two closely related bacteriophage, varphiCr30 and varphiCr35, are the first bacteriophage shown to mediate generalized transduction in Caulobacter crescentus. Unlike most other transducing phage, they are virulent and do not form any sort of lysogenic relationship with their host. However, they are rather inefficient at adsorption, so that transductants have a good chance of survival. The phage...
متن کاملUse of the Caulobacter crescentus genome sequence to develop a method for systematic genetic mapping.
The functional analysis of sequenced genomes will be facilitated by the development of tools for the rapid mapping of mutations. We have developed a systematic approach to genetic mapping in Caulobacter crescentus that is based on bacteriophage-mediated transduction of strategically placed antibiotic resistance markers. The genomic DNA sequence was used to identify sites distributed evenly arou...
متن کاملCell cycle constraints on capsulation and bacteriophage susceptibility
Despite the crucial role of bacterial capsules in pathogenesis, it is still unknown if systemic cues such as the cell cycle can control capsule biogenesis. In this study, we show that the capsule of the synchronizable model bacterium Caulobacter crescentus is cell cycle regulated and we unearth a bacterial transglutaminase homolog, HvyA, as restriction factor that prevents capsulation in G1-pha...
متن کاملRole of the flagellum in cell-cycle-dependent expression of bacteriophage receptor activity in Caulobacter crescentus.
The rate of adsorption of Caulobacter bacteriophage phi CbK to Caulobacter crescentus is dependent on the structural integrity of the flagellum. Cells lacking part or all of the flagellum because of either mutation or mechanical shear were defective in adsorption, and the extent of the defect in adsorption reflected the amount of flagellar structure missing. Maximal adsorption rates were also d...
متن کاملSpecific Assay for Differentiation in the Stalked Bacterium Caulobacter crescentus.
The bacterium Caulobacter crescentus carries out a distinct differentiation process during its life cycle. Cultures of the bacterium have been synchronized and an assay has been developed for monitoring the course of morphogenesis by the selective adsorption of radioactive RNA bacteriophage.
